Mw estimation

#1 Post by Giulia » 2017.05.08 18:18

Hello,

I am working with an elongated protein that behaves like a dimer in solution. I analyse the saxs profile using Primus and I calculated the Porod volume in order to determine the MW since the I0 does not give me the right estimation. The problem is that the MW expected is 160 kda but the Porod volume is~ 600 and consequently I have a MW of 330, double of what I m expecting. I calculated the Pr volume using both dataporod and SAXS Mow2 (same result). I am having the same situation also for a domain of the same protein that is 86 kDa but with the same analysis I am obtaining 160 kda, double the mass. The proteins have been studied by MALLS so I am sure about the MW. I am suspicious because in both cases I m obtaining double the expected mass.
Someone have an idea why is this happening?
Thanks a lot for the help
Giulia
